The Adverse Effect Profile of Acitretin in Children
TAKE-HOME MESSAGE
- The rates of clinical and laboratory adverse effects were analyzed in this retrospective case review of 174 children treated with acitretin in the UK. Laboratory adverse effects occurred in 22%, leading to cessation in 4% of the cohort. The most frequent laboratory abnormalities were increased triglycerides alone (10%), abnormal alkaline phosphatase alone (5.9%), and abnormal liver function tests and increased triglycerides (2.4%). In 3 children with ichthyosis, decreased bone density was noted but not routinely screened for. Diagnosis, demographics, age at starting acitretin, length of time on acitretin, and dosage were not significantly associated with rates of clinical adverse effects.

Abstract
The clinical benefit of acitretin has been amply demonstrated in the treatment of disorders of keratinization in childhood, particularly in psoriasis and ichthyosis. The adverse effects (AEs) of acitretin are well-studied in adults, and monitoring guidelines issued by the British Association of Dermatologists (BAD)1. However, AEs in childhood are less well-studied, particularly in non-psoriasis cohorts.
