Oral Fluconazole in Pregnancy Linked to Some Birth Defects, Not Others

A new study, published in The BMJ, adds to the evidence regarding use of oral fluconazole during pregnancy and risk for congenital malformations. 

Using a Medicaid database, researchers studied nearly 38,000 pregnancies exposed to oral fluconazole and 82,000 exposed to topical azoles during the first trimester (based on prescriptions dispensed). After multivariable adjustment, oral fluconazole was associated with significantly increased risk for musculoskeletal malformations (corresponding to 12 excess malformations per 10,000 exposed pregnancies), with the greatest risk seen at doses above 450 mg. Risks for the other primary outcomes — oral clefts and conotruncal malformations — did not differ significantly between the groups.
